REAGEN Scan Kit is a Mycoplasma detection kit based on TaqMan qPCR technology. It can detect more than 200 Mycoplasma, Ureaplasma, Mesoplasma, Spiroplasma, Acholeplasma and Phytoplasma, and does not cross-react with eukaryotic cells. The 2 x Premix master mix provided in the kit contains mycoplasma-specific FAM-MGB probe, internal control (Internal Control, IC) DNA and its specific ROX probe.

1. 2xPremix: 1 mL for 100 reactions
2. HS-Taq: 20 µL for 100 reactions
3. Positive control: 10 µL
* After receiving the goods, store at -20︒C; mix the enzyme with 2x premix before use, and store in aliquots and protected from light.
A. Sample preparation
Collect the cell culture medium that has been cultured for at least 2 days.
B. PCR reaction
1. Add 10 µL of 2xPremix premixed with HS-Taq enzyme to each qPCR reaction tube.
2. Add 2 µL of the cell culture medium or control sample to be tested, and add 8 µL of sterile water to a total volume of 20 µL.
3. Start the program on the computer and select dual-channel FAM-MBG and ROX-None.
4. Amplification reaction conditions: 95 °C pre-denaturation for 5 min; 95 °C 15 sec, 60 °C 30 sec, 45 cycles.
1. Save the data;
2. Read the fluorescence intensity of FAM and ROX channels, FAM is the fluorescence signal of mycoplasma, and ROX is the internal control fluorescence signal;
3. Judge the test results based on the amplification curve and Ct value.
